A few thoughts about observables and their developments in fluorescence bioimaging

Living matter provides major challenges for modelling. One avenue concerns defining a level of description, which can be experimentally addressed. The question of the observables is crucial with respect to this issue. In this lecture, I will recapitulate the state-of-the-art of the relevant observables, which have been made available over the years by chemists. I will more specifically focus on fluorescence imaging, which is presently (and will remain) central in the practice of biologists. In particular, I will introduce our recent developments aiming at providing access to highly multiplexed observations while overcoming several present limitations such as interference by ambient light or autofluorescence.
